What the fuck is fun about being killed by an invincible character 1000 times, until you eventually figure out the completely aribitrary weakness they've given the character. e.g. character is completely invincible, except that after they've attacked you 5 times, they go into the corner for a wank (5 shuffles), during which time they can be attacked.
It's even more fucking annoying, when there's no way to tell if you are actually damaging the character, or if you have to wait until a specific point to be able to damage them.
That said, some games do it pretty well. Like Half Life, that gives you a good introduction to some of the boss characters (Like when the scientist gets obliterated by the monster that tracks you by sound, before you have to encounter it)

Both Half Life and Half Life 2 do the does the 'boss' character fantasticly. A part of Half Life 2 episode 2 that i love is being chased in the Sandlion tunnels by the giant glowing bug (at this point in the game you dont really have much in terms of high powered weoponry to take it down) or when you come to the abandoned radio station and are attacked by several of those mini stalker/hunter things.
I think fable had some decent Boss fights (especially the Lost Chapter with the Dragon/Jack combo at the end)and im looking forward to Fable 2.
I like a tough end of game boss however. Cant wait for Aliens - Colonial Marines. Fighting a Queen was always fun in AvP 1 and 2 so interested to see how they do it in the new game.
